Bacteria play a vital role in life on Earth, inhabiting soil, water, plants, and animals.
For example, bacteria in the soil break down organic materials, recycling nutrients
back to the environment. Other bacteria live in and on our bodies and protect us
from disease. Some bacteria, however, can cause disease—these bacteria are
known as pathogens.
In order to minimize the risk of disease from bacteria, it is necessary to control their
growth in a variety of ways, including the use of antibiotics, disinfectants, and
antiseptics.
Under the proper conditions, bacteria can be grown in the laboratory. In this lab, you
will analyze the effects of various antimicrobial treatments on the growth of three
different bacteria—Staphylococcus aurenus, Haemophilus influenzae, and Streptococcus
pneumoniae.
The procedure below outlines the steps needed to grow bacteria and apply various
antimicrobial treatments in the lab. Navigate through this virtual lab by selecting the
arrows on the slider. Images and a description of each step are provided for you to
follow along.
Three separate petri dishes containing agar will be inoculated with three different
types of bacteria. Various antimicrobial agents will then be added to each dish. It is
your job to measure the bacterial growth around each antimicrobial agent to
determine the effectiveness of each treatment.

Inoculation
Inoculate the agar in the petri dish with bacteria from one of the test tubes.
Begin with test tube #1.
Test tube #1: Staphylococcus aurenus
Test tube #2: Haemophilus influenzae
Test tube #3: Streptococcus pneumoniae
Dip a sterile metal loop into the sample and streak it onto the agar plate in a
zig-zag pattern.

1. Define antibiotic, antiseptic, and disinfectant. What is the main difference
between the three types of microbial agents? Classify the seven treatments in
this lab as either an antibiotic, antiseptic, or disinfectant.
2. Use the results from your data table to create a clustered bar graph. Plot the type
of antimicrobial agent on the x-axis, and the size of bacterial growth inhibition (in
mm) on the y-axis. Use a legend to differentiate between bacteria.
3. Compare the bars on your graph to determine the effectiveness of various
antimicrobial agents on different bacteria. Which agents were effective at
stopping growth of all of the bacteria? Which agents were bacteria specific? How
do you know?
4. Describe the disease(s) caused by each of the three bacteria in this lab.
5. If you were a doctor treating a patient with pneumonia, which antibiotic would you
prescribe? Why?
6. Why is it important for antibacterial drugs to be bacteria-specific?
